This sequence change affects codon 835 of the POLE mRNA. It is a 'silent' change, meaning that it does not change the encoded amino acid sequence of the POLE protein. The c.2505C>T variant (also known as p.V835V), located in coding exon 22 of the POLE gene, results from a C to T substitution at nucleotide position 2505. This nucleotide substitution does not change the amino acid at codon 835. This nucleotide position is not well conserved in available vertebrate species. In silico splice site analysis predicts that this alteration may result in the creation or strengthening of a novel splice acceptor site. Based on the available evidence, the clinical significance of this variant remains unclear. -
