This sequence change replaces valine, which is neutral and non-polar, with alanine, which is neutral and non-polar, at codon 774 of the POLE protein (p.Val774Ala). This variant is present in population databases (rs771591846, gnomAD 0.01%). This variant has not been reported in the literature in individuals affected with POLE-related conditions. ClinVar contains an entry for this variant (Variation ID: 573332). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. RNA analysis performed to evaluate the impact of this missense change on mRNA splicing indicates it does not significantly alter splicing (Invitae). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -

The p.V774A variant (also known as c.2321T>C), located in coding exon 21 of the POLE gene, results from a T to C substitution at nucleotide position 2321. The valine at codon 774 is replaced by alanine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
