Three of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 2.8e-05 in 251486 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.2209A>G has been reported in the literature in two individuals affected with pancreatic cancer and breast cancer(Fujitani_2023, Guindalini_PMS2_2022), without strong evidence for causality. The p.T737A variant (also known as c.2209A>G), located in coding exon 20 of the POLE gene, results from an A to G substitution at nucleotide position 2209. The threonine at codon 737 is replaced by alanine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear. -
