This sequence change replaces histidine, which is basic and polar, with glutamine, which is neutral and polar, at codon 511 of the POLE protein (p.His511Gln). The p.H511Q variant (also known as c.1533C>G), located in coding exon 15 of the POLE gene, results from a C to G substitution at nucleotide position 1533. The histidine at codon 511 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
