This missense change has been observed in individual(s) with clinical features of POLE-related conditions (PMID: 26251183). ClinVar contains an entry for this variant (Variation ID: 405748). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt POLE protein function with a positive predictive value of 80%. The p.V460M variant (also known as c.1378G>A), located in coding exon 14 of the POLE gene, results from a G to A substitution at nucleotide position 1378. The valine at codon 460 is replaced by methionine, an amino acid with highly similar properties. This alteration has been detected in an individual diagnosed with cutaneous melanoma at age 29 and her father who was diagnosed with prostate cancer, colorectal cancer, and acute myeloid leukemia; additional relatives in this family were diagnosed with colorectal, esophageal, and prostate cancers (Aoude LG et al. Fam. Cancer, 2015 Dec;14:621-8). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
