This variant is present in population databases (rs150032060, gnomAD 0.03%). This missense change has been observed in individual(s) with endometrial cancer (PMID: 25505230, 34326862). ClinVar contains an entry for this variant (Variation ID: 405877). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt POLE protein function with a negative predictive value of 80%. Experimental studies have shown that this missense change does not substantially affect POLE function (PMID: 25228659). This variant has been reported in individual(s) with endometrial cancer (PMID: 25505230). To our knowledge, this variant has not been reported in the literature in individuals with POLE-related disease. The p.A428T variant (also known as c.1282G>A), located in coding exon 13 of the POLE gene, results from a G to A substitution at nucleotide position 1282. The alanine at codon 428 is replaced by threonine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
