Genes affected
POLE (HGNC:9177): (DNA polymerase epsilon, catalytic subunit) This gene encodes the catalytic subunit of DNA polymerase epsilon. The enzyme is involved in DNA repair and chromosomal DNA replication. Mutations in this gene have been associated with colorectal cancer 12 and facial dysmorphism, immunodeficiency, livedo, and short stature. [provided by RefSeq, Sep 2013]

Uncertain significance, criteria provided, single submitterclinical testingGenetic Services Laboratory, University of ChicagoNov 30, 2021DNA sequence analysis of the POLE gene demonstrated a sequence change, c.1015G>A, in exon 10 that results in an amino acid change, p.Asp339Asn. This sequence change has been described in the gnomAD database with a frequency of 0.11% in the African/African American subpopulation (dbSNP rs149029910). The p.Asp339Asn change affects a highly conserved amino acid residue located in a domain of the POLE protein that is known to be functional. The p.Asp339Asn substitution appears to be benign using several in-silico pathogenicity prediction tools (SIFT, PolyPhen2, Align GVGD, REVEL). This sequence change does not appear to have been previously described in individuals with POLE-related disorders. Due to insufficient evidences and the lack of functional studies, the clinical significance of the p.Asp339Asn change remains unknown at this time. -
