The c.737_753del17insA variant, located in coding exon 8 of the POLE gene, results from the deletion of 17 nucleotides and insertion of one nucleotide causing a translational frameshift with a predicted alternate stop codon (p.V246Efs*5). In silico splice site analysis predicts that this alteration will not have any significant effect on splicing. However, RNA studies have demonstrated that this alteration results in a splice defect; the clinical impact of this abnormal splicing is unknown at this time (Ambry internal data). Based on the available evidence, the clinical significance of this variant remains unclear. -
