The encoded protein is part of a large protein complex called ASCOM, which has been shown to be a transcriptional regulator of the beta-globin and estrogen receptor genes. This sequence change falls in intron 47 of the KMT2D gene. It does not directly change the encoded amino acid sequence of the KMT2D protein, but it affects a nucleotide within the consensus splice site of the intron. This variant is not present in population databases (ExAC no frequency). This variant has been shown to arise de novo in an individual affected with Kabuki syndrome (PMID: 24633898). Experimental studies have shown that this variant disrupts splicing, leading to the partial deletion of exon 48 and causing a downstream frameshift and subsequent premature stop codon (PMID: 24633898). In summary, this is a rare intronic change that has been seen in an affected individual and has been shown to disrupt splicing. However, without additional functional and/or genetic data, this variant has been classified as a Variant of Uncertain Significance. -
