Variant summary
Our verdict is Pathogenic. The variant received 12 ACMG points: 12P and 0B. PVS1PM2PP5_Moderate
The NM_000020.3(ACVRL1):c.18dupC(p.Arg7GlnfsTer31) variant causes a frameshift change involving the alteration of a non-conserved nucleotide. The variant was absent in control chromosomes in GnomAD project. Variant has been reported in ClinVar as Pathogenic (★). Synonymous variant affecting the same amino acid position (i.e. R7R) has been classified as Likely benign.

The c.18dupC pathogenic mutation, located in coding exon 1 of the ACVRL1 gene, results from a duplication of C at nucleotide position 18, causing a translational frameshift with a predicted alternate stop codon (p.R7Qfs*31). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
