The CDK4 c.626G>A (p.Arg209His) variant has been identified in the published literature in a reportedly healthy individual (PMID: 36243179 (2022)). The frequency of this variant in the general population, 0.000085 (3/35440 chromosomes in Admixed American subpopulation (Genome Aggregation Database, http://gnomad.broadinstitute.org)), is higher than would generally be expected for pathogenic variants in this gene. Analysis of this variant using bioinformatics tools for the prediction of the effect of amino acid changes on protein structure and function yielded predictions that this variant is benign. Based on the available information, we are unable to determine the clinical significance of this variant. -

The p.R209H variant (also known as c.626G>A), located in coding exon 4 of the CDK4 gene, results from a G to A substitution at nucleotide position 626. The arginine at codon 209 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
