The p.G15S variant (also known as c.43G>A), located in coding exon 1 of the CDK4 gene, results from a G to A substitution at nucleotide position 43. The glycine at codon 15 is replaced by serine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

This sequence change replaces glycine, which is neutral and non-polar, with serine, which is neutral and polar, at codon 15 of the CDK4 protein (p.Gly15Ser). This variant is present in population databases (no rsID available, gnomAD 0.0009%). This variant has not been reported in the literature in individuals affected with CDK4-related conditions. ClinVar contains an entry for this variant (Variation ID: 463466).
