Loss of function has been suggested as a mechanism of disease in this gene and has been reported in individuals with neurodevelopmental disorder (MONDO:0700092), HMGB1-related. The mechanism for brachyphalangy, polydactyly, and tibial aplasia/hypoplasia (MONDO:0012374), HMGB1-related is unclear, however, it has been reported that individuals with protein extension variants have this condition (PMIDs: 34164801, 36755093). (I) 0107 - This gene is associated with autosomal dominant disease. (I) 0205 - Variant is predicted to result in a truncated protein (premature termination codon is NOT located at least 54 nucleotides upstream of the final exon-exon junction) with less than 1/3 of the protein sequence affected.
