Variant summary: BRCA2 c.682-15C>T alters a nucleotide located at a position not widely known to affect splicing. Consensus agreement among computation tools predict no significant impact on normal splicing (TrAP). However, these predictions have yet to be confirmed by functional studies. The variant was absent in 241450 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.682-15C>T has been reported in the literature in individuals affected with prostate cancer without strong evidence for causality (Petrovics_2018). These report(s) do not provide unequivocal conclusions about association of the variant with Hereditary Breast And Ovarian Cancer Syndrome. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. ClinVar contains an entry for this variant (Variation ID: 219430).
