The missense variant c.905C>G(p.Thr302Ser) in BRCA2 gene has been submitted to ClinVar as a Variant of Uncertain Significance, but no details are available for independent assessment. It has not been reported in affected individuals. The p.Thr302Ser variant is observed in 0.002% alleles from individuals in gnomAD Exomes and is novel (not in any individuals) in 1000 Genomes. The amino acid Thr at position 302 is changed to a Ser changing protein sequence and it might alter its composition and physico-chemical properties The variant is predicted to be damaging by SIFT and the residue is conserved across species. The amino acid change p.Thr302Ser in BRCA2 is predicted as conserved by GERP++ and PhyloP across 100 vertebrates. For these reasons, this variant has been classified as Uncertain Significance. -
This sequence change replaces threonine with serine at codon 302 of the BRCA2 protein (p.Thr302Ser). The threonine residue is weakly conserved and there is a small physicochemical difference between threonine and serine. This variant is present in population databases (rs80359158, ExAC 0.02%). This variant has not been reported in the literature in individuals affected with BRCA2-related conditions. ClinVar contains an entry for this variant (Variation ID: 52737). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -

This missense variant replaces threonine with serine at codon 302 of the BRCA2 protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been detected in at least two individuals affected with breast or ovarian cancer (PMID: 33471991; Leiden Open Variation Database DB-ID BRCA2_007260, 33552952). This variant has been identified in 6/239016 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -
Hereditary cancer-predisposing syndrome Uncertain:2Benign:1
Missense variant in a coldspot region where missense variants are very unlikely to be pathogenic (PMID:31911673). -

The p.T302S variant (also known as c.905C>G), located in coding exon 9 of the BRCA2 gene, results from a C to G substitution at nucleotide position 905. The threonine at codon 302 is replaced by serine, an amino acid with similar properties. This alteration was identified in an individual diagnosed with breast and/or ovarian cancer (Kadri MSN et al. Front Oncol, 2020 Jan;10:568786). This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

Hereditary breast ovarian cancer syndrome Uncertain:1
This sequence change replaces threonine, which is neutral and polar, with serine, which is neutral and polar, at codon 302 of the BRCA2 protein (p.Thr302Ser). This variant is present in population databases (rs80359158, gnomAD 0.02%). This missense change has been observed in individual(s) with BRCA2-related conditions (PMID: 33552952). ClinVar contains an entry for this variant (Variation ID: 52737). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt BRCA2 protein function with a negative predictive value of 95%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
