Genes affected
BRCA2 (HGNC:1101): (BRCA2 DNA repair associated) Inherited mutations in BRCA1 and this gene, BRCA2, confer increased lifetime risk of developing breast or ovarian cancer. Both BRCA1 and BRCA2 are involved in maintenance of genome stability, specifically the homologous recombination pathway for double-strand DNA repair. The largest exon in both genes is exon 11, which harbors the most important and frequent mutations in breast cancer patients. The BRCA2 gene was found on chromosome 13q12.3 in human. The BRCA2 protein contains several copies of a 70 aa motif called the BRC motif, and these motifs mediate binding to the RAD51 recombinase which functions in DNA repair. BRCA2 is considered a tumor suppressor gene, as tumors with BRCA2 mutations generally exhibit loss of heterozygosity (LOH) of the wild-type allele. [provided by RefSeq, May 2020]

Pathogenic, criteria provided, single submitterclinical testingLabcorp Genetics (formerly Invitae), LabcorpJan 26, 2024This sequence change creates a premature translational stop signal (p.Asn319Lysfs*8) in the BRCA2 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in BRCA2 are known to be pathogenic (PMID: 20104584). This variant is present in population databases (rs756186207, gnomAD 0.006%). This premature translational stop signal has been observed in individual(s) with breast cancer (PMID: 12920083, 28724667). This variant is also known as c.951dupA and c.1179insA. ClinVar contains an entry for this variant (Variation ID: 52870). For these reasons, this variant has been classified as Pathogenic. -
Pathogenic, criteria provided, single submitterclinical testingLaboratory for Molecular Medicine, Mass General Brigham Personalized MedicineFeb 27, 2017The p.Asn319fs variant in BRCA2 has been reported in 1 Caucasian individual with breast cancer (Ozcelik 2003). It has also been identified in 1/8618 of East A sian chromosomes by the Exome Aggregation Consortium (ExAC, http://exac.broadins titute.org; dbSNP rs80359770). This variant is predicted to cause a frameshift, which alters the protein?s amino acid sequence beginning at position 319 and lea ds to a premature termination codon 8 amino acids downstream. This alteration is then predicted to lead to a truncated or absent protein. Heterozygous loss of f unction of the BRCA2 gene is an established disease mechanism in breast cancer. In addition, this variant was classified as Pathogenic on Sept 8, 2016 by the Cl inGen-approved ENIGMA expert panel (ClinVar SCV000300375.2). In summary, this va riant meets criteria to be classified as pathogenic for breast cancer in an auto somal dominant manner based upon the predicted impact to the protein. -
Pathogenic, criteria provided, single submitterclinical testingWomen's Health and Genetics/Laboratory Corporation of America, LabCorpSep 14, 2020Variant summary: BRCA2 c.956dupA (p.Asn319LysfsX8) results in a premature termination codon, predicted to cause a truncation of the encoded protein or absence of the protein due to nonsense mediated decay, which are commonly known mechanisms for disease. Truncations downstream of this position have been classified as pathogenic by our laboratory. The variant allele was found at a frequency of 8.7e-06 in 230430 control chromosomes. c.956dupA has been reported in the literature in multiple individuals affected with Hereditary Breast And Ovarian Cancer Syndrome (Ozcelik_2003, Lubinski_2004, Thirthagiri_2008, Mehta_2018). These data indicate that the variant is very likely to be associated with disease. Ten clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation. All laboratories classified the variant as pathogenic. Based on the evidence outlined above, the variant was classified as pathogenic. -

This variant has been reported in at least five unrelated individuals affected with breast or ovarian cancer (PMID: 18627636, 28724667, 30555256, 33471991; Leiden Open Variation Database DB-ID BRCA2_004164; Color internal data). Pathogenic, criteria provided, single submitterclinical testingAmbry GeneticsJun 05, 2024The c.956dupA pathogenic mutation, located in coding exon 9 of the BRCA2 gene, results from a duplication of A at nucleotide position 956, causing a translational frameshift with a predicted alternate stop codon (p.N319Kfs*8). This mutation has been reported in several individuals with hereditary breast and ovarian cancer from various ethnic backgrounds, including a Malaysian woman with breast cancer diagnosed at age 34 (Ozcelik H et al. J. Med. Genet., 2003 Aug;40:e91; Lubinski J et al. Fam Cancer, 2004;3:1-10; Thirthagiri E et al. Breast Cancer Res, 2008 Jul;10:R59; Sun J et al. Clin Cancer Res, 2017 Oct;23:6113-6119). Of note, this alteration is also designated as c.951dupA, 1184insA and 1179insA in published literature. In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
