In one study, this alteration was reported in 1/434 Nigerian breast cancer patients unselected for age of onset or family history (Fackenthal JD et al. Int. J. Cancer 2012 Sep; 131(5):1114-23). This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Four of five in-silico tools predict a benign effect of the variant on protein function. The variant was absent in 251138 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.1694C>T has been reported in the literature in at-least two individuals of African origin who were affected with Breast Cancer (example, Fackenthal_2012, Pal_2015), however it was also found in 1/2559 African American women, who are older than 70 years of age, and never had cancer (FLOSSIES database). These report(s) do not provide unequivocal conclusions about association of the variant with Hereditary Breast And Ovarian Cancer Syndrome. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Five clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation.
