This variant is present in population databases (no rsID available, gnomAD 0.006%). This variant has not been reported in the literature in individuals affected with BRCA2-related conditions. ClinVar contains an entry for this variant (Variation ID: 664262). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt BRCA2 protein function with a negative predictive value of 95%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
This sequence variant is a single nucleotide substitution (C>T) that results in an proline to leucine amino acid change at residue 655 in the BRCA2 protein. This variant is rare, and is absent (0/117974 alleles) from the ExAC database and a frequency of 0.000004224 (1/236724 alleles) in the gnomAD database. The variant was not found in any published reports or databases of known BRCA2 variants. A known benign variant (BRCA2:c.1964C>G,p.Pro655Arg) occurs at this site, but as leucine and arginine differ greatly in biochemical properties, it is not possible to say that this variant is likely to share similar properties. Thus, we consider this to be a variant of uncertain significance. -

The p.P655L variant (also known as c.1964C>T), located in coding exon 10 of the BRCA2 gene, results from a C to T substitution at nucleotide position 1964. The proline at codon 655 is replaced by leucine, an amino acid with similar properties. This alteration was identified in a cohort of 481 Chinese breast cancer patients with family history of breast and/or ovarian cancer (Wang J et al. Cancer Med, 2019 May;8:2074-2084). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

This missense variant replaces proline with leucine at codon 655 of the BRCA2 protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual affected with breast and/or ovarian cancer (PMID: 30982232) and in a breast cancer case-control meta-analysis in 2/60466 cases and 1/53461 unaffected individuals (PMID: 33471991; Leiden Open Variation Database DB-ID BRCA2_007911). This variant has been identified in 1/241642 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -
