This missense variant replaces glycine with valine at codon 1174 of the BRCA2 protein. Computational prediction suggests that this variant may not impact protein structure and function. To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual affected with ovarian cancer (PMID: 32211327). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -
PM2_Supporting, BP1_Strong c.3521G>T, located in exon 11 of the BRCA2 gene, is predicted to result in the substitution of Glycine by Valine at codon 1174, p.(Gly1174Val). This position is outside a (potentially) clinically important functional domain and, moreover, the SpliceAI algorithm predicts no significant impact on splicing (BP1_strong).It is not present in the population database gnomAD v2.1.1, non cancer dataset (PM2_supporting). To our knowledge, neither relevant clinical data nor well-stablished functional studies have been reported for this variant. In addition, the variant has been identified in the ClinVar database (1x likely benign, 2x uncertain significance), and BRCA Exchange database, (where it has not been reviewed yet), but it is not present in the LOVD database. The p.G1174V variant (also known as c.3521G>T), located in coding exon 10 of the BRCA2 gene, results from a G to T substitution at nucleotide position 3521. The glycine at codon 1174 is replaced by valine, an amino acid with dissimilar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt BRCA2 protein function with a negative predictive value of 95%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
