This variant has been reported in 10 individuals affected with breast and ovarian cancer from 6 families in Northern Spain (PMID: 23683081). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). Loss of BRCA2 function is a known mechanism of disease (clinicalgenome.org). Based on the available evidence, this variant is classified as Pathogenic.

The c.4030_4035delAATGATinsC pathogenic mutation, located in coding exon 10 of the BRCA2 gene, results from the deletion of 6 nucleotides (AATGAT) and insertion of one nucleotide (C), causing a translational frameshift with a predicted alternate stop codon (p.N1344Hfs*6). In one study, this mutation was identified in six breast and/or ovarian cancer families from a small, geographically isolated region in Northern Spain (Blay P et al. BMC Cancer, 2013 May;13:243). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
