This missense variant replaces lysine with glutamic acid at codon 1440 of the BRCA2 protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). Functional studies have shown this variant decreases affinity of BRCA2 for RAD51 in mammalian two-hybrid assays (PMID: 23071527). This variant has not been reported in individuals affected with hereditary cancer in the literature. This variant has been identified in 2/242658 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.

The p.K1440E variant (also known as c.4318A>G), located in coding exon 10 of the BRCA2 gene, results from an A to G substitution at nucleotide position 4318. The lysine at codon 1440 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.

This sequence change replaces lysine, which is basic and polar, with glutamic acid, which is acidic and polar, at codon 1440 of the BRCA2 protein (p.Lys1440Glu). This variant is present in population databases (rs80358668, gnomAD 0.007%). This missense change has been observed in individual(s) with clinical features of BRCA2-related conditions (PMID: 10923033). ClinVar contains an entry for this variant (Variation ID: 51631). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt BRCA2 protein function with a negative predictive value of 95%. Experimental studies have shown that this missense change affects BRCA2 function (PMID: 23071527). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
This sequence variant is a single nucleotide substitution (A>G) that results in a lysine to glutamic acid amino acid change at residue 1440 in the BRCA2 protein. This variant is rare; it is absent from the ExAC database and found at a frequency of 0.000008385 in the gnomAD database (2/238518 alleles). The residue is found in BRC repeat 3 (BRC3), and in a region required for the activation of polymerase H DNA polymerization activity, but there are no known pathogenic variants characterized in the immediate vicinity. Mammalian two-hybrid capture in vitro studies indicate the BRC3 domain with the K1440E variant has a significant ~60% decrease in affinity for RAD51 (PMID: 23071527). Due to the lack of well-established evidence, it is currently not possible to determine if this variant is pathogenic or benign; thus, it is a variant of uncertain significance.
