The p.R1677I variant (also known as c.5030G>T), located in coding exon 10 of the BRCA2 gene, results from a G to T substitution at nucleotide position 5030. The arginine at codon 1677 is replaced by isoleucine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

This missense variant replaces arginine with isoleucine at codon 1677 of the BRCA2 protein. Computational prediction suggests that this variant may not impact protein structure and function. To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual affected with pediatric leukemia (PMID: 26580448). This variant has been identified in 1/31370 chromosomes in the general population by the Genome Aggregation Database (gnomAD). This sequence change replaces arginine, which is basic and polar, with isoleucine, which is neutral and non-polar, at codon 1677 of the BRCA2 protein (p.Arg1677Ile). This variant is present in population databases (rs375304428, gnomAD 0.004%). This variant has not been reported in the literature in individuals affected with BRCA2-related conditions. ClinVar contains an entry for this variant (Variation ID: 220506). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt BRCA2 protein function with a negative predictive value of 95%.
