This changes the amino acid from a serine to a stop codon within coding exon 10. This mutation was detected in 2/8085 consecutive unselected Chinese breast cancer patients who underwent multi-gene panel testing. (Sun J et al. Clin Cancer Res, 2017 Oct;23:6113-6119). This mutation was also identified in two cohorts of women with ovarian cancer (Carter NJ et al. Gynecol Oncol, 2018 12;151:481-488; Rumford M et al. Sci Rep, 2020 02;10:3390). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay.
