The proline at codon 1702 is replaced by serine, an amino acid with similar properties. In one study, this alteration was detected in 1/102 breast or ovarian cancer probands from South India and was classified as a variant of uncertain significance by the study authors (Syamala V et al. J. Cancer Res. Clin. Oncol. 2007 Nov;133(11):867-74). Another study of a Sri Lankan cohort detected this alteration in 1/48 early onset breast cancer patients who also had a strong family history of breast cancer and 0/25 healthy controls without a personal or family history of breast cancer (De Silva S et al. Fam. Cancer. 2017 Jul;16:329-338). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
