The histidine at codon 1731 is replaced by tyrosine, an amino acid with similar properties. In a study of 1854 high-risk breast and ovarian cancer families in Italy, this alteration was detected in one family (Azzollini J et al. Eur. J. Intern. Med., 2016 Jul;32:65-71). This variant has also been identified in multiple individuals diagnosed with breast cancer (Borg A et al. Hum. Mutat., 2010 Mar;31:E1200-40; Su Y et al. Front Genet, 2021 Nov;12:674094). This variant has also been reported in an individual diagnosed with breast cancer; however this individual was also positive for a different pathogenic BRCA2 mutation (Santonocito C et al. Cancers (Basel), 2020 May;12:). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |

The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.5191C>T has been reported in the literature in individuals affected with Hereditary Breast and Ovarian Cancer (Borg_2010, Azzollini_2016, Zanella_2017). One of these publications reported this variant in a proband who carried another pathogenic mutation, however the exact identity of this co-occurring variant was not specified (Azzollini_2016). To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Three clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation.
