The p.Y1762D variant (also known as c.5284T>G), located in coding exon 10 of the BRCA2 gene, results from a T to G substitution at nucleotide position 5284. The tyrosine at codon 1762 is replaced by aspartic acid, an amino acid with highly dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

The BRCA2 c.5284T>G (p.Tyr1762Asp) variant has been reported in the published literature in individuals with breast cancer (PMID: 35264596 (2022)) and a Lynch syndrome associated cancer and/or polyps (PMID: 25980754 (2015)). This variant has also been reported to be located in a region of the BRCA2 gene that is tolerant to missense sequence changes (PMID: 31911673 (2020)). The frequency of this variant in the general population, 0.000008 (2/250334 chromosomes (Genome Aggregation Database, http://gnomad.broadinstitute.org)), is uninformative in the assessment of its pathogenicity. Analysis of this variant using bioinformatics tools for the prediction of the effect of amino acid changes on protein structure and function yielded predictions that this variant is benign. Based on the available information, we are unable to determine the clinical significance of this variant. -
