The p.Q1782E variant (also known as c.5344C>G or 5572C>G), located in coding exon 10 of the BRCA2 gene, results from a C to G substitution at nucleotide position 5344. The glutamine at codon 1782 is replaced by glutamic acid, an amino acid with some highly similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. To date, this alteration has been detected with an allele frequency of approximately 0.003% (greater than 105,000 alleles tested) in our clinical cohort. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of p.Q1782E remains unclear. -

ClinVar contains an entry for this variant (Variation ID: 142990). This variant is not present in population databases (ExAC no frequency). This sequence change replaces glutamine with glutamic acid at codon 1782 of the BRCA2 protein (p.Gln1782Glu). The glutamine residue is weakly conserved and there is a small physicochemical difference between glutamine and glutamic acid. -
