The p.V2174A variant (also known as c.6521T>C), located in coding exon 10 of the BRCA2 gene, results from a T to C substitution at nucleotide position 6521. The valine at codon 2174 is replaced by alanine, an amino acid with similar properties. This alteration was detected in 2/1197 patients with breast cancer who underwent genetic testing (Abdel-Razeq H et al. Front Oncol, 2022 Mar;12:673094). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear. -
This missense variant replaces valine with alanine at codon 2174 of the BRCA2 protein. Computational prediction suggests that this variant may not impact protein structure and function. To our knowledge, functional studies have not been reported for this variant. This variant has been reported in two individuals affected with breast cancer (PMID: 35402282). This variant has been identified in 1/242468 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -

Variant summary: The c.6521T>C (p.Val2174Ala) in BRCA2 gene is a missense variant involves a non-conserved nucleotide and 3/5 in silico tools predict benign outcome, however no functional studies supporting these predictions were published at the time of evaluation. The variant is located outside of any known functional domain or repeat. The c.6521T>C is absent from the control population dataset of ExAC (~120414chrs tested), but is present at a low frequency in gnomAD (0.000004; 1/237750 chrs tested). This frequency does not exceed the maximal expected allele frequency for a disease causing allele in BRCA2 gene (0.000175). To our knowledge, the variant has not been reported in affected individuals via published reports but is cited as VUS by reputable databases/clinical laboratories. Taken together, the variant was classified as VUS, until new information becomes available. -
