The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.6829C>T has been reported in the literature in at least one family with a suspected hereditary predisposition to breast/ovarian cancer, however without strong evidence for causality (e.g., Machackova_2019). This report does not provide unequivocal conclusions about association of the variant with Hereditary Breast And Ovarian Cancer Syndrome. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 31409081, 25348012). Four ClinVar submitters (evaluation after 2014) have cited the variant with conflicting assessments: three submitters classified the variant as uncertain significance, and one submitter classified the variant as likely benign.
