This sequence change replaces proline, which is neutral and non-polar, with threonine, which is neutral and polar, at codon 2283 of the BRCA2 protein (p.Pro2283Thr). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with ovarian cancer (PMID: 23683081). ClinVar contains an entry for this variant (Variation ID: 462417). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt BRCA2 protein function with a negative predictive value of 95%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -

It has not been reported in large, multi-ethnic general populations (http://gnomad.broadinstitute.org). In the published literature, the variant has been reported in individuals with a personal or family history of breast and/or ovarian cancer (PMIDs: 33471991 (2021) , 31853058 (2020), 23683081 (2013)), as well as in healthy individuals (PMID: 33471991 (2021)). Analysis of this variant using bioinformatics tools for the prediction of the effect of amino acid changes on protein structure and function yielded predictions that this variant is damaging. Based on the available information, we are unable to determine the clinical significance of this variant. -
Hereditary cancer-predisposing syndrome Uncertain:1
The p.P2283T variant (also known as c.6847C>A), located in coding exon 11 of the BRCA2 gene, results from a C to A substitution at nucleotide position 6847. The proline at codon 2283 is replaced by threonine, an amino acid with highly similar properties. This alteration was observed in 1 of 256 families from northern Spain with breast and/or ovarian cancer; but the variant did not segregate with disease in the family, which had two case of ovarian cancer (Blay P et al. BMC Cancer 2013; 13:243). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
