This changes the amino acid from a glutamic acid to a stop codon within coding exon 13. This alteration has been previously reported in a breast/ovarian cohort as well as an ancestrally diverse cohort unselected for cancer history (Olfson E et al. PLoS ONE, 2015 Sep;10:e0135193; Rebbeck TR et al. Hum. Mutat., 2018 May;39:593-620). This alteration has also been identified in a large, worldwide study of BRCA1/2 mutation positive families (Rebbeck TR et al. Hum Mutat, 2018 05;39:593-620). Another study detected this mutation in 0/3030 pancreatic cancer cases and 2/123136 population controls (Hu C et al. JAMA, 2018 06;319:2401-2409). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay.
