This variant inserts 1 nucleotide in exon 14 of the BRCA2 gene, creating a frameshift and premature translation stop signal. This variant is expected to result in an absent or non-functional protein product. To our knowledge, this variant has not been reported in individuals affected with hereditary cancer in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). Loss of BRCA2 function is a known mechanism of disease (clinicalgenome.org). Based on the available evidence, this variant is classified as Pathogenic. -
The c.7177dupA pathogenic mutation, located in coding exon 13 of the BRCA2 gene, results from a duplication of A at nucleotide position 7177, causing a translational frameshift with a predicted alternate stop codon (p.M2393Nfs*19). This mutation (referred to as c.7171_7172insA) has previously been reported in an individual diagnosed with early onset breast cancer (de Juan Jiménez I et al. Fam Cancer. 2013 Dec;12:767-77). This alteration was also identified in a large, worldwide study of BRCA1/2 mutation-positive families (Rebbeck TR et al. Hum Mutat. 2018 05;39:593-620). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
