The p.H2415R variant (also known as c.7244A>G), located in coding exon 13 of the BRCA2 gene, results from an A to G substitution at nucleotide position 7244. The histidine at codon 2415 is replaced by arginine, an amino acid with highly similar properties. This alteration was identified in an individual diagnosed with ocular melanoma (Scott RJ et al. Int J Cancer, 2002 Nov;102:188-91). This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
This missense variant replaces histidine with arginine at codon 2415 of the BRCA2 protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). Splice site prediction tools suggest that this variant may not impact RNA splicing. To our knowledge, functional studies have not been performed for this variant. This variant has been reported in an individual affected with ocular melanoma (PMID: 12385017). This variant has been identified in 2/251274 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
