Variant summary: BRCA2 c.7438T>G (p.Leu2480Val) results in a conservative amino acid change in the encoded protein sequence. Four of four in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 8e-06 in 251152 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.7438T>G has been reported in the literature in individuals affected with/undergoing testing for breast cancer (example, Singh_2018, Torrorey-Sawe_2020). These reports do not provide unequivocal conclusions about association of the variant with Hereditary Breast And Ovarian Cancer Syndrome. At-least one computational method reported this variant with a probabilistic likelihood ratio of neutral (Karchin_2008). To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Three clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation. All laboratories classified the variant as uncertain significance. Based on the evidence outlined above, the variant was classified as uncertain significance. -

The p.L2480V variant (also known as c.7438T>G), located in coding exon 14 of the BRCA2 gene, results from a T to G substitution at nucleotide position 7438. The leucine at codon 2480 is replaced by valine, an amino acid with highly similar properties. This variant has been identified in multiple individuals diagnosed with breast and/or ovarian cancer (Singh J et al. Breast Cancer Res Treat, 2018 Jul;170:189-196; Torrorey-Sawe R et al. Front Genet, 2020 Mar;11:170). This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear. -

This variant is present in population databases (rs80358965, gnomAD 0.01%). This missense change has been observed in individual(s) with breast and/or ovarian cancer (PMID: 29470806, 32231682). ClinVar contains an entry for this variant (Variation ID: 52331). An algorithm developed specifically for the BRCA2 gene suggests that this missense change is likely to be tolerated (PMID: 19043619). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
