This variant has been identified in cohorts of ethnic Chinese individuals with indications for genetic testing for hereditary breast and ovarian cancer (Bhaskaran SP et al. Int J Cancer, 2019 Aug;145:962-973; Gao X et al. Hum Mutat, 2020 Mar;41:696-708). This nucleotide position is highly conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will weaken the native splice acceptor site. RNA studies have demonstrated that this alteration results in abnormal splicing in the set of samples tested (Fraile-Bethencourt E et al. PLoS Genet, 2017 Mar;13:e1006691; Mesman RLS et al. Genet Med, 2020 Aug;22:1355-1365; Ambry internal data). In addition, this alteration failed to rescue the loss of Brca2 activity in an assay of cell survival in murine embryonic stem cells (Mesman RLS et al. Genet Med, 2020 Aug;22:1355-1365). Alterations that disrupt the canonical splice site are expected to cause aberrant splicing, resulting in an abnormal protein or a transcript that is subject to nonsense-mediated mRNA decay.
