Five of five in-silico tools predict a benign effect of the variant on protein function. The variant was absent in 249656 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.8225A>G has been reported in the literature in individuals affected with breast cancer (example, Manguoglu_2003). These report(s) do not provide unequivocal conclusions about association of the variant with Hereditary Breast And Ovarian Cancer Syndrome. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Two clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation. All laboratories classified the variant as uncertain significance citing overlapping evidence utilized in the context of this evaluation. This alteration has been reported in a cohort of Turkish individuals with presumed inherited predisposition to breast cancer, male breast cancer, and early onset breast cancer (Manguoglu AE et al. Hum Mutat. 2003 Apr; 21(4):444-5). This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis.
