This variant is denoted BRCA2 c.8527A>G at the cDNA level, p.Asn2843Asp (N2843D) at the protein level, and results in the change of an Asparagine to an Aspartic Acid (AAT>GAT). Using alternate nomenclature, this variant would be defined as BRCA2 8755A>G. This variant has been observed in at least one individual with breast cancer (Carney 2010). BRCA2 Asn2843Asp was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, suggesting it is not a common benign variant in these populations. Since Asparagine and Aspartic Acid differ in some properties, this is considered a semi-conservative amino acid substitution. BRCA2 Asn2843Asp occurs at a position that is not conserved and is located in the DNA binding domain (Yang 2002). In silico analyses are inconsistent regarding the effect this variant may have on protein structure and function. Based on currently available evidence, it is unclear whether BRCA2 Asn2843Asp is a pathogenic or benign variant. We consider it to be a variant of uncertain significance. -
Hereditary cancer-predisposing syndrome Uncertain:2
The p.N2843D variant (also known as c.8527A>G), located in coding exon 19 of the BRCA2 gene, results from an A to G substitution at nucleotide position 8527. The asparagine at codon 2843 is replaced by aspartic acid, an amino acid with highly similar properties. This alteration has been reported in individuals diagnosed with breast and/or ovarian cancer (Carney ME et al. Hawaii Med J, 2010 Nov;69:268-71; Jasiewicz A et al. Hered Cancer Clin Pract, 2022 Apr;20:12). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
This missense variant replaces asparagine with aspartic acid at codon 2843 of the BRCA2 protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual affected with breast cancer and an individual affected with ovarian cancer (PMID: 21218378, 35382848). This variant has been identified in 1/251166 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -

This sequence change replaces asparagine, which is neutral and polar, with aspartic acid, which is acidic and polar, at codon 2843 of the BRCA2 protein (p.Asn2843Asp). This variant is present in population databases (no rsID available, gnomAD 0.0009%). This missense change has been observed in individual(s) with breast cancer (PMID: 21218378). ClinVar contains an entry for this variant (Variation ID: 233431). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt BRCA2 protein function with a negative predictive value of 95%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
