This variant has been identified in 3/282626 chromosomes in the general population by the Genome Aggregation Database (gnomAD). Invitae Evidence Modeling incorporating data from in vitro experimental studies (PMID: 33609447) indicates that this missense variant is not expected to disrupt BRCA2 function with a negative predictive value of 95%. Experimental studies have shown that this missense change does not substantially affect BRCA2 function (PMID: 29884841, 35736817). Three of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 4e-06 in 251218 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.8545A>G has been reported in the literature in individuals affected with Hereditary Breast And Ovarian Cancer Syndrome (Pal_2013, Alvarez_2017). These reports do not provide unequivocal conclusions about association of the variant with Hereditary Breast And Ovarian Cancer Syndrome. Co-occurrence with a pathogenic variant has been reported (BRCA1 c.3817C>T, p.Gln1273Ter, BIC database), providing supporting evidence for a benign role. At least two publications report experimental evidence evaluating an impact on protein function. These results showed no damaging effect of this variant (Guidugli_2018, Hu_2022). These results showed no damaging effect of this variant. The following publications have been ascertained in the context of this evaluation (PMID: 29088781, 29394989, 38417439, 26287763).
