The p.L2865F variant (also known as c.8595A>T), located in coding exon 19 of the BRCA2 gene, results from an A to T substitution at nucleotide position 8595. The leucine at codon 2865 is replaced by phenylalanine, an amino acid with highly similar properties. One study detected this alteration in 1/512 Polish women with familial and/or early onset breast and/or ovarian cancer (Kluska A et al. BMC Med Genomics, 2015 May;8:19). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
