This position is inside a (potentially) clinically important functional domain and is predicted to have a significant impact on splicing by Splice AI as it predicts that the variant disrupts the canonical acceptor site (SpliceAI-AcceptorLoss:0,29) and generates a novel cryptic splice acceptor site downstream of the canonical site (SpliceAI-AcceptorGain: 0,28), that could cause the in-frame skipping of first 54 bases of exon 23 (r.8954_9007del; p.Val2985_Glu3002del) (PP3).It is not present in the population database gnomAD v2.1.1, non cancer dataset (PM2_supporting). To our knowledge, neither relevant clinical data nor well-stablished functional studies have been reported for this variant.
