The p.C3233F variant (also known as c.9698G>T), located in coding exon 26 of the BRCA2 gene, results from a G to T substitution at nucleotide position 9698. The cysteine at codon 3233 is replaced by phenylalanine, an amino acid with highly dissimilar properties. This nucleotide position is well conserved in available vertebrate species. In silico splice site analysis predicts that this alteration may result in the creation or strengthening of a novel splice acceptor site. An RNA minigene assay demonstrated this alteration resulted in an acceptor gain (Acedo A et al. Hum Mutat, 2015 Feb;36:210-21).This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear. -
Hereditary breast ovarian cancer syndrome Uncertain:1
This sequence change replaces cysteine, which is neutral and slightly polar, with phenylalanine, which is neutral and non-polar, at codon 3233 of the BRCA2 protein (p.Cys3233Phe). This variant is present in population databases (rs398122620, gnomAD 0.003%). This variant has not been reported in the literature in individuals affected with BRCA2-related conditions. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt BRCA2 protein function. Studies have shown that this missense change does not affect mRNA splicing (PMID: 25382762). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
