ATP7B (HGNC:870): (ATPase copper transporting beta) This gene is a member of the P-type cation transport ATPase family and encodes a protein with several membrane-spanning domains, an ATPase consensus sequence, a hinge domain, a phosphorylation site, and at least 2 putative copper-binding sites. This protein is a monomer, and functions as a copper-transporting ATPase which exports copper out of the cells, such as the efflux of hepatic copper into the bile. Alternate transcriptional splice variants, encoding different isoforms with distinct cellular localizations, have been characterized. Variant summary: ATP7B c.1621G>A (p.Glu541Lys) results in a conservative amino acid change located in the Heavy metal-associated domain (IPR006121) of the encoded protein sequence. Five of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 0.0001 in 252882 control chromosomes. This frequency is not significantly higher than estimated for a pathogenic variant in ATP7B causing Wilson Disease (0.0001 vs 0.0054), allowing no conclusion about variant significance. c.1621G>A has been reported in the literature in at least one individual affected with Wilson Disease who was recorded in the Wilson Disease Mutation Database, which has been subsequently cited by others (e.g. Kenney_2007, Coffey_2013, Collet_2018). This report does not provide unequivocal conclusions about association of the variant with Wilson Disease. At least one publication reports experimental evidence evaluating an impact on protein function and found that the variant had no effect on the binding of ATP7B to COMMD1, however, this does not allow convincing conclusions about the overall variant effect (de Bie_2007). The following publications have been ascertained in the context of this evaluation (PMID: 23518715, 30097039, 17919502, 17680703). ClinVar contains an entry for this variant (Variation ID: 92386).
