FLVCR2 (HGNC:20105): (FLVCR choline and putative heme transporter 2) This gene encodes a member of the major facilitator superfamily. The encoded transmembrane protein is a calcium transporter. Unlike the related protein feline leukemia virus subgroup C receptor 1, the protein encoded by this locus does not bind to feline leukemia virus subgroup C envelope protein. The encoded protein may play a role in development of brain vascular endothelial cells, as mutations at this locus have been associated with proliferative vasculopathy and hydranencephaly-hydrocephaly syndrome. This sequence change creates a premature translational stop signal (p.Ile353Serfs*9) in the FLVCR2 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in FLVCR2 are known to be pathogenic (PMID: 20206334, 20690116). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with FLVCR2-related conditions. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. For these reasons, this variant has been classified as Pathogenic. -
