To our knowledge, this variant has not been reported in the literature. This variant is reported in 0.0018% of alleles in individuals of European (Non-Finnish) descent in gnomAD. However, a different missense variant impacting the same amino acid residue, p.Ala553Thr, has been reported in homozygous and compound heterozygous individuals with a hypothyroidism phenotype (Abramowicz et al. 1997. PubMed ID: 9185526; Park et al. 2004. PubMed ID: 14725684). Taken together, while we suspect that the c.1657G>T (p.Ala553Ser) variant could be pathogenic, at this time we interpret its clinical significance as uncertain due to the absence of conclusive functional and genetic evidence. - |
