Variant summary: GALC c.1592G>C (p.Arg531Pro) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant was absent in 248936 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.1592G>C in individuals affected with Krabbe Disease and no experimental evidence demonstrating its impact on protein function have been reported. A different variant affecting the same codon has been classified as pathogenic by our lab (c.1592G>A, p.Arg531His), supporting the critical relevance of codon 531 to GALC protein function. ClinVar contains an entry for this variant (Variation ID: 623356). Based on the evidence outlined above, the variant was classified as uncertain significance. -
