DICER1 (HGNC:17098): (dicer 1, ribonuclease III) This gene encodes a protein possessing an RNA helicase motif containing a DEXH box in its amino terminus and an RNA motif in the carboxy terminus. The encoded protein functions as a ribonuclease and is required by the RNA interference and small temporal RNA (stRNA) pathways to produce the active small RNA component that represses gene expression. This protein also acts as a strong antiviral agent with activity against RNA viruses, including the Zika and SARS-CoV-2 viruses. Alternative splicing results in multiple transcript variants. This sequence change creates a premature translational stop signal (p.Tyr1874*) in the DICER1 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 49 amino acid(s) of the DICER1 protein. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with DICER1-related conditions. ClinVar contains an entry for this variant (Variation ID: 933131). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -

The p.Y1874* variant (also known as c.5622C>G), located in coding exon 26 of the DICER1 gene, results from a C to G substitution at nucleotide position 5622. This changes the amino acid from a tyrosine to a stop codon within coding exon 26. Premature stop codons are typically deleterious in nature, however, this stop codon occurs at the 3' terminus of DICER1, is not expected to trigger nonsense-mediated mRNA decay, and removes only the last 49 amino acids of the protein. The exact functional impact of these removed amino acids is unknown at this time. -
