Hereditary cancer-predisposing syndrome Uncertain:2
Uncertain significance, criteria provided, single submitter | curation | Sema4, Sema4 | May 24, 2021 | - - |
Uncertain significance, criteria provided, single submitter | clinical testing | Ambry Genetics | Feb 03, 2023 | The p.V1260I variant (also known as c.3778G>A), located in coding exon 20 of the DICER1 gene, results from a G to A substitution at nucleotide position 3778. The valine at codon 1260 is replaced by isoleucine, an amino acid with highly similar properties. This alteration was identified in an individual diagnosed with thyroid cancer (Canberk S et al. Eur Thyroid J, 2021 Feb;9:296-303). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
not provided Uncertain:1
Uncertain significance, criteria provided, single submitter | clinical testing | GeneDx | Jul 06, 2023 | Not observed at significant frequency in large population cohorts (gnomAD); In silico analysis supports that this missense variant does not alter protein structure/function; Observed in an individual with papillary thyroid carcinoma (Canberk et al., 2021); This variant is associated with the following publications: (PMID: Canberk2022[article], 33718253) - |
DICER1-related tumor predisposition Uncertain:1
Uncertain significance, criteria provided, single submitter | clinical testing | Labcorp Genetics (formerly Invitae), Labcorp | Jan 19, 2024 | This sequence change replaces valine, which is neutral and non-polar, with isoleucine, which is neutral and non-polar, at codon 1260 of the DICER1 protein (p.Val1260Ile). This variant is present in population databases (rs763425076, gnomAD 0.002%). This missense change has been observed in individual(s) with papillary thyroid carcinoma (PMID: 33718253). ClinVar contains an entry for this variant (Variation ID: 412085). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt DICER1 protein function with a negative predictive value of 80%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. - |
