This changes the amino acid from a tyrosine to a stop codon within coding exon 20. A different nucleotide substitution (c.3675C>G) resulting in the same stop codon (p.Y1225*) has been previously identified in the tumor of an individual diagnosed with pineal blastoma at age 5y; however, specimen was not available for germline confirmation (de Kock L et al. Acta Neuropathol., 2014 Oct;128:583-95). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. - |
