This sequence change replaces histidine, which is basic and polar, with proline, which is neutral and non-polar, at codon 1124 of the DICER1 protein (p.His1124Pro). This variant is present in population databases (rs534996867, gnomAD 0.01%). This variant has not been reported in the literature in individuals affected with DICER1-related conditions. ClinVar contains an entry for this variant (Variation ID: 571626). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt DICER1 protein function with a negative predictive value of 80%. The p.H1124P variant (also known as c.3371A>C), located in coding exon 20 of the DICER1 gene, results from an A to C substitution at nucleotide position 3371. The histidine at codon 1124 is replaced by proline, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
