The genetic deficiency of IVD results in an accumulation of isovaleric acid, which is toxic to the central nervous system and leads to isovaleric acidemia. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. ClinVar contains an entry for this variant (Variation ID: 2440925). This variant is also known as p.Arg21Leu. This missense change has been observed in individual(s) with isovaleric acidemia (PMID: 17576084; Invitae).
